Cynthia McFadden Lectures Clinton on Running for President: Don't You Have an 'Obligation?

As if comparing Hillary Clinton to?Thomas Jefferson?wasn't hyperbolic enough, ABC's Cynthia McFadden on Wednesday practically begged the Secretary of State to run for president. The?Nightline?co-anchor lobbied Clinton, lecturing her about the "obligation" she has to "break through that glass ceiling." Yet, the reporter could only manage the most gentle probing into the issue of the terrorist attack in Libya.

McFadden pressed the Democrat to run in 2016, asking the question no less than four times. If it appeared, the journalist wondered, "that you might be the person who could actually break through that glass ceiling and become the first female president of this country, would you feel a certain obligation to seize that mantel?" [MP3 audio?here.] Any answer other than yes seemed not good enough.

She pushed, "But there's never been a woman who really had a credible chance. And it looks as if you might just be that person. And I know how seriously you take commitment and obligation."

The journalist pleaded, "...Are we up to maybe?"

Finally, after quizzing Clinton about her health, McFadden got to the issue of dead Americans in Libya. She charitably described the Secretary of State's evasive testimony before Congress this way: "In her testimony, Clinton pulled no punches."

McFadden's first question wasn't really a query at all. Citing Clinton's line about "what difference does it make" over how the attack occurred, the anchor sympathized, "It seemed as though you lost your temper at the hearing."

She then asked the Secretary if she "regretted" that statement and whether, "you stand by what you said?"

What kind of a question is that? Would Clinton say no? How about pressing her on the details of the attack?"

ABC donated 15 minutes over three programs (Nightline,?World News?and?Good Morning America)?to Clinton. At least McFadden could offer some truly hard questions. She opened the segment by pretending it would be this way, declaring that "No topic was off-limits, from the most contentious global issues to the most intensely personal ones."

Topics may not have been "off limits," but they certainly were framed in a friendly way. For more on this interview,?go here.

A partial transcript of the January 30 segment is below:

12:49am EST

CYNTHIA MCFADDEN: She has met with world leaders 1,700 times in the past four years, but as of Friday, Hillary Clinton will no longer be secretary of state. Today, Massachusetts Senator John Kerry was confirmed as her successor.?I've interviewed Secretary Clinton nearly a dozen times and I've never seen her so relaxed or upbeat. We sat down for her last television interview in office. No topic was off-limits, from the most contentious global issues to the most intensely personal ones. She's been Secretary of State for four years. Visited 112 countries. More than any of her predecessors. Logged almost a million miles.?But it's where she's going next that has everybody talking. Let's just say this isn't the first time the question has come up.?In Moscow three years ago, you told me, "I have absolutely no interest in running for president, none." You're never going to run for president again?"

HILLARY CLINTON [file footage] : I have absolutely no interest in running for president again. None, none.

MCFADDEN: Two years ago you said the exact same thing in Australia. [File footage.] You said talk about the future. You said you're not running for president in 2012 or 2016 or 2020. [Cut to present day.] And yet in the past few days, a PAC called Ready for Hillary has been launched. Can you still say with a straight face that there's no way you would consider running for president?

CLINTON: Sitting here now, that is certainly what I believe, and I am, you know, still the Secretary of State, so I'm not in politics. I'm going to be focusing on my philanthropy, my charities, my writing and speaking. So I am looking forward to having something resembling a normal life again.

MCFADDEN:?And yet, are we up to maybe?

CLINTON [Laughs]: That's very good, Cynthia. Well, of course, of course, I am flattered and honored. I didn't even know about some of these things that are happening now. I don't know how else to say it, but I am going to get back into my life again. This is going to be new for me. I don't know how I'm going to react to it, to be honest.

MCFADDEN: When you conceded defeat in the primary, you made a famous speech in which you said that there were 18 million cracks in the glass ceiling.

CLINTON: Although we weren't able to shatter that highest, hardest glass ceiling this time, thanks to you, it's got about 18 million cracks in it.

MCFADDEN:?If in the course of the next couple years it appears, as it does appear right now, that you might be the person who could actually break through that glass ceiling and become the first female president of this country, would you feel a certain obligation to seize that mantel?

CLINTON: I do want to see that glass ceiling shattered. I don't think it has to be any particular person.

MCFADDEN:?But there's never been a woman who really had a credible chance. And it looks as if you might just be that person. And I know how seriously you take commitment and obligation.

CLINTON: Right. But I'm not making any commitments or obligations because I do take them seriously.

...

MCFADDEN:?What else goes with the territory? Something Clinton is certainly familiar with? harsh criticism and tough questions. She was called before Congress last week and grilled over what she has called the low point in her tenure, the security lapse in Benghazi, Libya, that led to the death of four Americans, including Ambassador Chris Stevens. In the first days, the controversy reached a boiling point over this.

RICE: There was a violent protest. Violent protest outside our embassy.

MCFADDEN: U.S. Ambassador to the U.N. Susan Rice's explanation that the murders were the result of an anti-Muslim video. That turned out to be false.?It was a pre-planned terrorist attack. In her testimony, Clinton pulled no punches.

CLINTON: We didn't know that. With all due respect, the fact is we had four dead Americans. Was it because they had protests, or because guys out for a walk and they decided to kill some Americans. What difference at this point does it make? It is our job to figure out what happened and do everything question to prevent it from ever happening again, senator.

MCFADDEN: And today, she doubled down.?It seemed as though you lost your temper at the hearing.

CLINTON: When someone tries to put it into a partisan lens, when they focus not on the fact that we had such a terrible event happening with four dead Americans, but instead, what did somebody say on a sunday morning talk show? That to me is not in keeping with the seriousness of the issue and the obligation we all have as public servants.

MCFADDEN: But do you regret "what difference at this point does it make?"

CLINTON: No, because I think that asking questions about talking points for a Sunday morning talk show, is really missing the point.?I believe in transparency. I said let the chips fall where they may. Put it all out there. And I don't want that to be politicized.

MCFADDEN: So you stand by what you said?

CLINTON: Absolutely.

Fog delays 3rd round at Torrey Pines

SAN DIEGO (AP) ? Tiger Woods is going to have to wait to pursue another win at Torrey Pines.

A thick fog shrouded the course along the Pacific bluffs on Saturday and essentially wiped out the entire day at the Farmers Insurance Open. Woods, a six-time winner of this tournament, had a two-shot lead and never even bothered coming to the golf course. Three players completed one hole ? and that was after a three-hour delay.

Players were to resume the third round Sunday morning and go as long as daylight allowed, and then finish Monday.

And that's a best-case scenario.

In a bizarre twist, tour officials were hopeful of rain and a little wind Sunday morning, two elements that most golfers dread. That's what is needed, however, to keep the fog away from Torrey Pines and allow the tournament to resume.

"When Mother Nature doesn't want you to play, you can't play," said Mark Russell, the tour vice president of competition.

They did just about everything else.

Lucas Glover warmed up three times, at one point passed the time with a little trickery. He lined up two balls in the direction of the range, and hit them with a wedge so that one ball went straight in the air, and Jerry Kelly took a baseball swing with an inverted club and made contact.

Through four tournaments this year, the PGA Tour already had had its share of weather problems. This will be the second tournament that doesn't finish on the scheduled day. The Tournament of Champions at Kapalua didn't even start until Monday, the day it was supposed to end, because of 40 mph gusts. It had to be reduced to 54 holes and was completed on Tuesday.

Woods was at 11-under 133 and didn't need to come to the course with all the delays because he was in the last group with Billy Horschel and Casey Wittenberg. Horschel spent part of his day getting advice through text messages on how to play with Woods.

He'll get to see plenty of Woods over the next two days.

Russell said there would not be a 54-hole cut for 87 players ? typically there is a cut when the field is more than 78 players on Saturday. Instead, they will take a short lunch break and go back out, racing time when fog allows. The groups will stay the same until the tournament is over.

"I think the coolest thing that can come from it is that I think it looks like we're going to be paired together for 36 holes," Horschel said. "So it's a pretty cool thing that's going to happen. It's just going to be a fun day tomorrow and Monday. So we'll see how it goes."

Horschel also is among 17 players who are entered in the qualifier Monday to get into the Phoenix Open, which starts next week. Russell said those players have been given the latest tee times for the qualifier ? the latest is 1:36 p.m. MST ? with hopes of getting there.

Saturday began with a 30-minute delay that soon stretched into three hours, and the sun broke through the clouds as Hunter Mahan, Robert Garrigus and John Mallinger teed off on the 10th hole at the South Course. Five minutes later, the horn sounded. Because it was for weather, they were able to finish the hole.

Mahan's caddie took a picture of the 11th tee, a 216-yard par 3, and the visibility was no more than 50 yards.

It never got any better.

"That little bit of sun we did get today was overpowered by just the saturation of the atmosphere, so we just couldn't overcome it today," said Stewart Williams, the tour's meteorologist.

Defending champion Brandt Snedeker, seven shots out of the lead, was among those who had the longest day. He was up at 5 a.m., arrived in darkness, warmed up in the fog and never hit a shot.

"It's tough when you prepare one day, and then it's one of those days where preparation is going to be kind of thrown off," he said. "But we have these days from time to time. We kind of know what to do to keep your mind fresh and ready to go. You realize it's a different week, and could be a Monday finish now and be ready for that."

Woods won in 2005 at Torrey Pines when fog interrupted the tournament, though it was completed on Sunday. The last time this event was shortened to 54 holes because of weather was in 1998.

Russell said the Farmers Insurance Open will go 72 holes. He just couldn't promise when 84 players could get to the 37th hole of the tournament. Without rain, there was the chance of more fog Sunday morning.

"If we say we're not going to play because we might have fog in the morning, and we didn't have fog, we'd have tremendous problems," he said. "So we're going to schedule it, and hopefully, we won't have fog and we can play golf. That's all we can do. If Mother Nature doesn't cooperate, we don't have very much control over that. We're going to be out here, ready to play, and see what happens."

That's assuming they can see at all.

Report: Japan to air ultra-high-def TV in 2014

11 hrs.

TOKYO???The Japanese government is set to launch the world's first 4K TV broadcast in July 2014, roughly two years ahead of schedule, to help stir demand for ultra high-definition televisions, the Asahi newspaper reported on Sunday without citing sources.

The service will begin from communications satellites, followed by satellite broadcasting and ground digital broadcasting, the report said.

The 4K TVs, which boast four times the resolution of current high-definition TVs, are now on sale by Japanese makers including Sony, Panasonic and Sharp. Other manufacturers include South Korea's LG Electronics.

Japan's Ministry of Internal Affairs and Communications had aimed to kick-start the 4K TV service in 2016. That has been brought forward to July 2014, when the final match of the 2014 football World Cup is set to take place in Brazil, the Asahi report said.

In Japan, the development of super high-definition 8K TVs is in progress, and the Ministry of Internal Affairs and Communications plans to launch the test 8K TV broadcast in 2016, two years ahead of schedule, it said.

Illinois Driver's Licenses For Undocumented Immigrants Law To Be Signed Sunday

SPRINGFIELD, Ill. -- As Illinois becomes the fourth and most populous state to issue driver's licenses to undocumented immigrants, there are still nagging concerns that the measure doesn't have enough safeguards to avoid the identity fraud and other pitfalls faced by the three other states with similar laws.

Backers of the proposal, who tout it as a public-safety measure, argue that required facial recognition technology is reliable enough to prevent fraud, but opponents point to hundreds of fraudulent cases in New Mexico, Washington and Utah after those states began giving undocumented immigrants permission to drive. Illinois will not require applicants to be fingerprinted, for fear that would discourage immigrants from applying.

"How many people would apply for this document knowing that fingerprints will be going to (federal authorities)? Probably not all that many," said Fred Tsao, policy director of the Illinois Coalition for Immigrant and Refugee Rights, a driving-force behind the measure.

Illinois Gov. Pat Quinn is expected to sign the measure that grants driving privileges to undocumented immigrants Sunday. Proponents say it will allow an estimated 250,000 individuals unlawfully residing in the state to apply for a three-year temporary driver's license and require them to get training and insurance.

The licenses will be like those already issued to certain foreign-born, legal visitors. Under the new law, applicants will be photographed at a driver services facility, and their photo will be entered into the state's facial recognition database - like the rest of Illinois' licensed drivers - to verify their identity.

But the other states' driving programs for undocumented immigrants have been abused. New Mexico and Washington both issue licenses, while Utah issues a permit.

An Associated Press investigation last year found a striking pattern in New Mexico, suggesting immigrants tried to game the system to obtain a license. In one instance, 48 foreign-born individuals claimed to live at a smoke shop in Albuquerque to fulfill a state residency condition. New Mexico does not have a fingerprinting requirement, though it asks applicants to show two proofs of state residency.

Authorities also busted a fraud ring last year that forged documents for undocumented immigrants to use after driving from as far as Illinois and North Carolina to obtain a New Mexico license. Gov. Susana Martinez has vowed for years to repeal the decade-old measure, but the legislature has repeatedly rejected such efforts.

Washington's requirements attracted national attention when Jose Antonio Vargas, a Pulitzer Prize-winning journalist and then-Washington license-holder, revealed his undocumented immigration status in an essay for the New York Times Magazine in 2011. Vargas chronicled how he obtained his license. State authorities conducted an investigation that revealed Vargas did not reside at the address he stated in his application, and canceled his license a month after his essay was published.

Utah issues three different driving privilege cards: one for U.S. citizens and permanent residents, another for legal visitors and a third for undocumented immigrants. Utah's permit for undocumented immigrants is not valid for identification. Illinois' law will follow suit.

Utah's Republican-controlled legislature amended the state's law in 2011 to require undocumented immigrants to be fingerprinted, and mandates that the state notify U.S. Immigration and Customs Enforcement if an applicant's fingerprint check yields a felony on record. If the individual applying has a misdemeanor warrant outstanding, the state must notify the agency that is seeking the person's arrest.

That kind of information-sharing between state and immigration authorities worries Illinois' immigrant-rights advocates, like Tsao, who pushed for the legislation without a fingerprinting requirement. They say fingerprinting could deter potential licensees from applying for fear of being identified and deported.

Local law enforcement officials argue in favor of fingerprinting.

"We could see if they have committed a crime; it could be a crime in another state or it could be a crime in their home country," said John Kennedy, executive director of the Illinois Association of Chiefs of Police.

The Illinois secretary of state's office has vehemently defended its facial recognition database as highly sophisticated and accurate. The program uses an algorithm to match more than a dozen facial features that are not easy to alter, such as eye sockets and sides of the mouth.

"The integrity of our driver's license system is a priority," said Henry Haupt, a spokesman for the office.

The bill's sponsor, Rep. Edward Acevedo, a Chicago Democrat, said state roads will be safer because undocumented immigrants will receive training and be tested before obtaining a license. They also will be required to purchase insurance, an aspect that would save millions for currently insured drivers, Acevedo said.

Tsao's organization estimates uninsured undocumented immigrant drivers cause $64 million in damage claims each year, an expense currently covered by increased premiums.

Real Politix ? Crucial Advice For Anyone Investing In The Stock Market

TIP! Instead of purchasing a few costly stocks, think about buying a larger amount of affordable ones. These stocks are a lot easier to manage and sell.

Whether you?re an experienced trader or just starting out, everyone can benefit from stock market advice. Other than selling high and buying low, there are many different tips you can use to generate income. Read this article so that you can make the most money possible from the stock market.

TIP! Don?t ignore other opportunities just because you are invested in stocks. You can also invest in mutual funds, art, real estate, and bonds.

Diversifying your portfolio is much more complex than buying a few different stocks from various sectors. Besides that, not every element of your strategy needs to be incorporated into every investment opportunity. However, it helps if you can create a group of stocks that represent different sectors, using many other selection criteria as you are comfortable with.

TIP! If the goals of your portfolio are for maximum long term profits, you need to have stocks from various different industries. Not every sector will do well in any given year.

When analyzing a stock, look at its value, not its price. Is this stock intended to be held for a while? If the price is lower than normal, find out why before you invest, so you know beforehand if it is a good investment or not. Don?t just buy a cheap stock in the hopes that it will improve over time.

TIP! Have you considered paper trading? This is good practice without losing any actual money. This involves imaginary money, but with investment techniques which may be applied in the real market.

An important part of investing is re-evaluating your stock portfolio periodically, such as every quarter. The reason for that is the economy is changing frequently. Some industries will advance, while others will gradually die out. A wise financial investment of one year ago may be a poor financial investment today. You therefore need to track your portfolio and make changes as needed.

TIP! Trading stocks online is a good way to save money. Online firms can offer reduced prices that are not provided by standard brokerage firms.

Practice with a fake account before using actual money in the market. You don?t even need software to practice. Just choose a stock that you?d buy and track its current value. Watch how the stock fluctuates over a long time frame. This way, you can see how your judgement plays out without causing yourself any loss of money.

TIP! Find an investment service to subscribe to that you can rely on. One great stock market service will serve you well.

Before you buy any stock, you should be very clear on your goals and where this purchase fits in. It may be that you want to build and strengthen your portfolio, or you may be seeking out a low risk way to obtain income. No matter the case, it is important to create a strategy to get you to accomplish specific goals.

TIP! Sound portfolios can generate returns in the area of 8 percent, while terrific ones may bring 15 or 20 percent. It?s possible to break that barrier as well.

Keep in mind that when you purchase stocks, you buy ownership. Keep the company?s reputation in mind when selecting the stocks you buy. You have to do your own research of the company so that you make a wise choice to avoid losing an investment.

TIP! You should invest in large companies at first. If you?re a beginner, start with lower risk or low beta stocks.

Keep your plan simple if you?re just beginning. When you first start out it can seem hard to diversity, yet if you keep applying yourself and read as much as you can then you should have no problem succeeding. That one piece of advice might save you a lot of money over time.

TIP! If you are knowledgeable enough to do your own research, you may want to look into getting an online broker. The fees to trade and commissions on these online brokers are much cheaper that a discount or full service brokerage.

Before going to a broker, you should do some background research to make sure you can trust them with your money. If you take a little time to investigate the organization and understand their business practices, you will help to protect yourself against investment fraud.

TIP! Invest in industries you know. Some of the best investors such as Peter Lynch and Warren Buffet made most of their profits by investing in industries that they had knowledge in.

Researching companies and learning about their historical profit margins, key executives, reputation and other financial trends can help you make better investment decisions. Rather than getting your information from word of mouth, ensure you are remaining informed using excellent sources. Remembering the advice you learned here can ensure that you get the most profit from your trades.

Home Decor Can Either Ruin Or Save Your Marriage, Apparently

We've seen home decor do a lot of things for people. It can relax you, make you happy and even have an effect on your self-image. But can it save your marriage? According to Lauren Machling of the Wall Street Journal, perhaps it can.

In a recent article, Machling speaks of the problems that often arise in a marriage when furnishings of both partners are combined during cohabiting. She says rooms become designated as "his" and "hers" and pieces of furniture start to become known as "yours" and "mine" with a wedge being driven between the two -- if there can't be a compromise.

And according to Jay Lebow, a professor of psychology and a marital therapist and researcher at the Family Institute at Northwestern University in Chicago, it's things like fighting over lamps and rugs that can make one person feel less adequate, especially if it involves one moving into another's home. "The person who was living there all along feels hurt every time something is moved or changed. And the person who moved in feels like they've lost their voice," he told the newspaper.

But Machling says that doesn't have to be the way. With books like "Luxurious Minimalism" by photographer Fritz von der Schulenburg and writer Karen Howes, newlyweds can find ways to combine their two styles and compromised. The designs in the book, Machling writes, "reflect a certain ideal?one maintained by a couple who, while equally strong-willed, have learned to make room for each other's visual style."

Of course, you can never really tell. For instance, Machling spoke with author Jackie Collins, who, after two failed marriages, refuses to share a space with a man for fear of losing her glamourous, art deco style. "I don't think I ever could do it again. I have very distinctive tastes and I'm not going to concede to anybody," she said.

Head over the the Wall Street Journal to read the full article, and comment to weigh in: Can a relationship be affected by one's furnishings?

Weird News: Man Hits 100 MPH On Way To Booty Call | YourTango

man being arrested
Would you break the law for your booty call?

No friend with benefit is worth endangering innocent lives, right?

Rule Number One of the booty call game: don't let them think about it. Thinking with anything other than your baby-producing parts is a recipe for someone not answering the door. If you think, you end up using the internet to self-abuse.?

More from YourTango: Love Bytes: It's OK To Break Off An Engagement

Per the Chicago Tribune, a local man very much knows that getting there is 99% of the battle. In a rush to get to the domicile of a ladyfriend, he was clocked going 111 MPH and failed to brake for a both stop sign and a red light. After getting pulled over, the 21-year old man explained to the officers that he was merely breaking laws and endangering lives so that he could have sex with a woman whom he was not romantically attached to.

There is no doubt that the fuzz was sympathetic to his plight but hitting triple digits and blowing through red things (stop signs/ traffic lights) sort of ties their hands. Throw in trace amounts of reefer and you may have to text that woman to break into her in-case-of-emergency ?marital aid? kit.

And to add insult to blue balls, the man was sentenced to five days of medium labor picking up trash or washing cop cars. The man's next 18 months will feature probation and random drug tests. The potential booty call could not be reached for comment but, if I know the nature of these things, she probably watched an episode of Girls and went to bed after not hearing from him for 30 minutes.

This is the only reason I live in a city with plentiful cabs. Hopefully, the next time this young man has a booty call waiting, he'll obey traffic laws, invite her to his place or use Skype, like a gentleman. If I've heard it once, I've heard it a million times, amorous young men are a danger to themselves and everything around them and this administration needs to do something about it.

In similarly sad news, a Missouri man drove to a police precinct looking for a little comfort from this recent breakup and instead received a DWI, according to?Patch.com.

White House Petition Demands Cellphone Unlocking Remain Legal

Stan Schroeder

A petition submitted to the White House demands the Librarian of Congress to rescind his recent decision which removed the unlocking of cellphones from the exceptions to the Digital Millenium Copyright Act (DMCA).

Due to the Librarian's decision, the unlocking of cellphones effectively becomes illegal in the U.S. on Jan. 26.

"As of January 26, consumers will no longer be able unlock their phones for use on a different network without carrier permission, even after their contract has expired," reads the petition. "Consumers will be forced to pay exorbitant roaming fees to make calls while traveling abroad. It reduces consumer choice, and decreases the resale value of devices that consumers have paid for in full."

Created by "S. K." from San Francisco, CA, the petition needs to reach 100,000 signatures before February 23 in order to get a response from the White House ? the White House recently raised the threshold from 25,000 signatures. At the time of this writing, 3,121 people have signed the petition.

"We ask that the White House ask the Librarian of Congress to rescind this decision, and failing that, champion a bill that makes unlocking permanently legal," the petition text concludes.

Cheesy Chicken Sausage Pasta Bake

Do you guys watch the Bachelor? Or the Bachelorette? I mean this is taking my reality TV addiction to a whole new level but I figured I?d come clean. I?m an on and off again Bachelor kinda gal. I only watch it so I can read and actually understand what my friend Zack is talking about on his blog. Do you read it? Lost Angeles? Zack is an awesome writer, and an even better friend. He?s also married to one of my best friends in the whole wide world so even if I didn?t love him, I?d still be stuck with him forever :) I started a few seasons back, Ben?s season, I mean he went to our?high school, his sister is a friend of mine, and I figured it was as good a time as any to start.

Anyways, every Tuesday Zack writes this recap on the Bachelor. Unless he?s traveling, then it?s on Wednesday (rude). It?s something I look forward to at the end of each show because well, it?s hysterical. You should read it. And watch the Bachelor so it makes sense. I feel like one day Zack is going to be on the After the Final Rose special show or something to ask the?contestants?questions and make things really?awkward. It would be brilliant. He?s also a total fab football sports writer, but if you know me, unless Tim Riggins is involved, I don?t understand football.

So we decided to celebrate our mutual love for the worlds worst TV show as Zack like to call it, aka, the Bachelor and create a little dinner and drink party. Zack made cocktails and I made dinner. It?s what I do best. I decided to go all out.?Cheesy Chicken Sausage Pasta Bake. I?m fairly certain not one girl on the show this season would actually eat this because I?ve decided that none of them eat. Ever. But whatever, we do, so we can indulge.

It?s heavy on the carbs. Loaded with sausage and cream. And topped with a decadent amount of cheese. It?s indulgent to say the least. And it?s so perfect for the Bachelor. Zack also refers to it as,?carb-heavy, slut and glut comfort food recipe, which is kind of perfect if you ask me.

It?s basically super easy to make, feeds a boat load of people, and doubles as kick ass leftovers if you have any left after your Bachelor viewing party. So you should totally make this next Monday! And then tweet or instagram us pics?with the hashtag?#GZbachmeal

It?s going to be a party :)

For the tiny Daubenton's bat, the attractions of family life seem to vary more with altitude than with the allure of the opposite sex.

For more than a decade, a team led by Professor John Altringham from the University of Leeds' School of Biology has studied a population of several hundred bats along a 50-km stretch of the River Wharfe. They monitored roosts in Ilkley and Addingham, upstream in the market town of Grassington and higher still in the villages of Kettlewell and Buckden.

The researchers found that all Daubenton's bats in nursery roosts in lowland areas of Wharfedale during the spring and summer were females and their offspring.

Male bats were mostly restricted to a windier, Heathcliff-like existence in roosts at the top of the Dales.

But the researchers were surprised to find a small oasis of cohabitation in Grassington, sandwiched between the bustle of the women-only childrearing in the lowlands and the more relaxed lives of the bachelors in the highlands.

Professor Altringham said: "Low down the dale, the females appear not to tolerate males and we assume they won't let them in the roost. They don't want anything to do with them. High in the dales, all the roosts are bachelor pads. But in the middle, at Grassington, males and females live togetherthe social structure changes with the environment"

"One possible reason for not finding males low down the valley could be that the mothers just want to avoid competing with males for food. It takes a lot of insects to make the milk needed to feed their young," Professor Altringham said.

"But it is also possible that the males choose not to roost with the females. When you look at the nursery colony in Ilkley, mothers and pups often have a lot of ectoparasites like ticks and mites. In a warm, crowded nursery, parasites can thrive, especially if there's less time for good personal hygiene. Parasites not only make life uncomfortable but can affect a bat's health. The males that live by themselves are usually very clean in their bachelor pads, so you can understand why they might not want to move in," he added.

At Grassington, which is deep in the Yorkshire Dales National Park but not as high as Buckden and Kettlewell, the bats have a completely different social structure. Both male and female bats live with the young throughout the spring and summer in roosts in the stonework of the old Dales bridges and in holes in ash trees.

"Females may roost as high up the dale as Grassington because they have these warm, cuddly males to bunk up with. This way, females use less energy keeping warm and babies grow faster," Professor Altringham said. "In these marginal conditions, they may just tolerate a few males to keep them warm. Otherwise they kick them out. Why do the males co-habit if they are going to get parasites all over them? Well, that may be down to the usual answer: sex."

Although male and female Daubenton's bats usually live apart throughout the spring and summer, they meet when they begin flying to caves in late summer.

Professor Altringham said: "In and around these caves the bats gather in huge numbers to mate, in a behaviour known as swarming. This is clubbing for bats, with males displaying to females in lengthy acrobatic chases. As winter closes in, these caves will ultimately be their hibernation sites.

"There are nearly 2,000 cave entrances and hundreds of kilometres of cave passages in the Dales and these attract bats from all over Yorkshire, Lancashire, Cumbria and beyond for mating and hibernation. The males in Grassington may be giving themselves the opportunity to mate with the females late in the summer before they even get to the caves."

The researchers have built up a detailed picture of social and sexual behaviour by genotyping hundreds of individuals. The evidence gathered from this supports the theory that the Grassington males enjoy an advantage in mating.

"At Grassington, most of the fathers of bats born there spent the summer with the females. If we look at pups in Addingham and Ilkley, their dads were males caught when swarming at caves. So, as well as two different mating systems, you have distinct social groupings. A bachelor from Buckden is always a bachelor from Buckden. He doesn't pop down to Grassington to visit the females in the summer. His only option seems to be to go clubbing in the autumn," Professor Altringham said.

The Daubenton's bat, named after the 18th Century French naturalist Louis-Jean-Marie Daubenton, is widespread across the United Kingdom and specialises in hunting insects over water. Full-grown adults weigh only 7 to 12 grams, but they can live for 20 years or more.

"These bats are the size of a shrew but have a very different lifecycle. A shrew typically spends its entire life in a few metres of hedgerow, eats and breeds with a ferocious intensity, for a year if it is lucky, and then dies. In contrast, these bats lead a complex life over a huge area and females produce only one pup a year," Professor Altringham said. "This makes bats particularly vulnerable to the problems of habitat fragmentation and climate change."

Synthetic corkscrew peptide kills antibiotic-resistant Gram-negative bacteria

Friday, January 25, 2013

An engineered peptide provides a new prototype for killing an entire category of resistant bacteria by shredding and dissolving their double-layered membranes, which are thought to protect those microbes from antibiotics.

The synthetic peptide was effective in lab experiments against antibiotic-resistant Gram-negative bacteria, which cause a variety of difficult-to-treat, potentially lethal infections such as pneumonia and sepsis.

The team led by scientists at The University of Texas MD Anderson Cancer Center reported its findings online in advance of print this week at the Proceedings of the National Academy of Science.

"The antibiotic pipeline against multidrug-resistant Gram-negative problem pathogens is a major unmet need in contemporary medicine; as such, our new antimicrobial agent holds immediate promise," said co-senior author Wadih Arap, M.D., professor in MD Anderson's Department of Genitourinary Medical Oncology and the David H. Koch Center.

Arap, Renata Pasqualini, Ph.D., also a co-senior author, professor in genitourinary medical oncology and the Koch center, and colleagues have previously constructed peptide combinations that are in development against cancer and white fat cells.

"The prototype introduced here as an antibiotic candidate has a unique mechanism of action and translational applications readily identified," Pasqualini said.

Gram-negative bacteria that are highly resistant to existing treatments include E. coli, Acinetobacter baumanii, Pseudomonas aeruginosa, and kebsiella pneumonia. These infections are often present in health care settings and most threatening to people with weakened immune systems.

The spiral peptide called KLAKLAKKLAKLAK acts against bacteria by puncturing their lipid bilayer membranes and has only low toxicity toward mammalian cells. These antimicrobial peptides, however, are subject to routine destruction by host enzymes or those generated by the microbe. Combating that effect by increasing the dose heightens both toxicity to other cells and cost.

D- KLAKLAKKLAKLAK destroys microbes, biofilms

Arap, Pasqualini and colleagues engineered a version of KLAKLAKKLAKLAK to use in their combination therapies but had not tested the peptide alone as an antibiotic.

The peptide is made of L-amino acids, the building blocks of life, which makes them vulnerable to destruction. The researchers synthesized a peptidomimetic ? a version of the peptide using D-amino acids with a reversed peptide sequence, making it more durable.

In a series of lab experiments, the researchers found that D-KLAKLAKKLAKLAK:

  • Kills a variety of strains of E. coli, A. baumanii and P. aeruginosa, including multi-drug resistant strains.
  • Works against Gram-negative bacteria at all phases of growth, including dormant cells that are prone to become resistant.
  • Causes dose-dependent damage to the bacterial membrane resulting in its dissipation and cell death.
  • Specifically disrupts lipids found in Gram-negative bacteria membranes while not affecting membranes in eukaryotic cells ? cells with the nucleus and other structures enclosed in separate membranes found in mammals and other non-microbial life.
  • Works in combination with the antibiotic piperacillin at lower doses to kill bacteria.
  • Eliminates biofilms, layers of combinations of microbes that adhere to surfaces and provide an ideal setting for bacterial growth.

Next step: Animal model experiments

Arap and Pasqualini note that developing D- KLAKLAKKLAKLAK as a drug will next require experiments in animal models of sepsis and other infections to further gauge the peptide's effectiveness and side effects.

In their cancer and anti-obesity research, the D-peptide is used with targeting agents to hit specific cells. Large preclinical studies in mice, rats and monkeys showed low toxicity at treatment-level concentrations. Their cancer drug in a first-in-human phase I clinical trial revealed side effects that were predictable, dose-dependent and reversible. Even so, toxicity may differ when it's used against bacterial infections.

The peptide was not effective against Gram-positive bacteria, which have thicker cell walls but are generally more vulnerable to antibiotics and the immune system than are Gram-negative bacteria. Gram-positive bacteria include those that cause anthrax, tuberculosis, strep throat and such treatment-resistant infections as Staphylococcus aureus.

Gram-negative bacteria, which have thinner membranes but are generally more resistant to antibiotics or immune system attack, also include those that cause typhoid fever, cholera, gonorrhea, syphilis and lyme disease.

Microsoft's 2Q earnings dip despite Windows 8 lift

SAN FRANCISCO (AP) ? Microsoft's latest quarterly earnings slipped, even as the world's largest software maker showed modest progress adjusting to a shift away from the personal computers that have been its financial foundation for decades.

The results announced Thursday are the first to include Windows 8. The program is a dramatic overhaul of the Microsoft Corp. operating system that powers most PCs. Windows 8 came out Oct. 26 with slightly more than two months left in Microsoft's fiscal second quarter.

Microsoft is counting on Windows 8 to help the company extend its franchise into tablet computers while still reaping revenue from a new breed of PCs. The redesigned software displays applications in a mosaic of interactive tiles instead of a staid menu. It can be controlled by touching on a display screen, as well as the traditional method of using a keyboard and a mouse.

Although the Windows 8 sales haven't been as impressive as investors hoped, revenue in Microsoft's Windows division climbed 24 percent from the previous year. That includes sales that had to be deferred from earlier quarters because the purchases were made before Windows 8's release.

When Windows 8 finally hit the market, Microsoft also unveiled its own tablet computer, Surface, as a showcase for a streamlined version of the operating system. Microsoft didn't immediately disclose how many Surface devices were sold in the October-December period.

"I don't think they want to provide that because it won't be impressive," technology analyst Patrick Moorhead said. More details about the Surface could emerge Thursday evening when Microsoft management is scheduled to review the quarterly results with analysts on a conference call.

BGC Financial analyst Colin Gillis said he still has faith in Microsoft, saying the company "has multiple revenue streams that are still very nice businesses."

"I kind of like the Windows segment," he said, adding that the 24 percent growth was "a little stronger than expected."

But another Microsoft division that includes another big moneymaker ? the company's Office suite of software ? didn't fare as well. Revenue in the Office division declined 10 percent, a shortfall that may have spooked some investors. Analyst Josh Olson of Edward Jones believes many of Microsoft's corporate customers may have held off on buying Office because a new version of that program is scheduled to come out early this year.

Microsoft earned $6.4 billion, or 76 cents per share, during the final three months of the year. That was down 4 percent from $6.6 billion, or 78 cents per share, a year earlier.

The company's total revenue rose 3 percent from last year to $21.5 billion.

The earnings were a penny above the average estimate of analysts surveyed by FactSet, while the total revenue fell below analysts' projections by about $100 million.

Microsoft's stock shed 57 cents, or 2.1 percent, to $27.06 in Thursday's extended trading, after the release of results. The stock has remained stuck below its price before Windows 8 and Surface came out, signaling that investors aren't pleased with the products' performance so far.

"This is going to be a 'show-me' year for Microsoft, and there was nothing to really shout about in this quarter," Olson said. "So what we are seeing is sort of a shrug from investors."

Besides debuting Windows and Surface during the most recent quarter, Microsoft also released a new version of its operating phone for smartphones.

Moorhead called the Surface and the new software line-up the "keys to Microsoft's future."

If Microsoft's revamped software for tablets and smartphones catches on, it would help the company overcome a downturn in PC sales, which has reduced licensing revenue during the past year. Worldwide PC shipments fell 3.5 percent last year, marking the industry's first annual decline since 2001, according to the research firm Gartner Inc.

Despite Microsoft's high hopes and an elaborate marketing campaign, Windows 8 appears to have gotten off to a tepid start. Technology reviews have panned the software as too confusing and cumbersome to navigate, and none of the hundreds of devices running on Windows 8 emerged as a breakout hit during the holiday season.

A big chunk of Microsoft's Windows revenue in the holiday-season quarter came from sales that were made before the new operating system's release. Excluding revenue that had been deferred from previous quarter, Windows revenue increased 11 percent from the same 2011 period.

The Surface also hasn't been able to mount a significant challenge to Apple Inc.'s trend-setting iPad, Amazon.com Inc.'s Kindle Fire, Samsung Electronics Co.'s Galaxy or Google Inc.'s Nexus devices. Surface, which runs on a streamlined version of Windows 8, is meant to showcase how well the software works on a tablet. A version of Surface running the full version of Windows 8 comes out Feb. 9.

Gillis said Windows 8 is Microsoft's attempt to solve a disruption in the market that's taking place because of tablets.

"It's too early to declare it a success or a failure," he said.

"The sentiment on the PC market is just too negative," Gillis added, referring to the death knells that have been ringing through much of the technology industry during the past year. "Yes, there are disruptions going on but we still sell close to a million PCs a day."

Reiterating information released earlier this month, Microsoft said it has licensed more than 60 million copies of Windows 8. That puts the redesigned system on the same early sales trajectory as its predecessor, Windows 7, after it came out in 2009. It's unclear how many of the devices that have licensed Windows 8 are still sitting on store shelves.
